Hi Doug and santhosh,

Thanks alot for sharing your point of view. we now indeed asked our collaborators to give some more of their slides so we can do some regular IHCs to confirm that this tissue is the problem.
As soon as we have done that i'll share the results with the forum.

Hi Kuehla, another thing you can try is to do an IHC with the conjugated antibody in both pH6 and pH9. you will see some background due to the metals but you can still clearly see the specific signal from the antibody. We have experience with the 236A/E7 clone for IHC/IF in which the antibody works in both pH6 and pH9. we use a very standard method of boiling for 10' in the microwave in either citrate (pH6) or tris/edta buffer (pH9) and cooling down the slides for about 1h. We have the same experience.

1mmx1mm takes about 1h50m-2h at 200hz, 500x500 about 25m (which makes most sense if you would have 0,6mm cores).

Dear Matt, I assume you solved this months ago but in case anyone else wonders in the future. In our experiments we do a 5 minute fixation with cold metanol directly after cutting and then use both surface and IC antibodies together. We have a number of IC markers and they all work properly.
